RNA immunoprecipitation (RIP) is similar to chromatin immunoprecipitation (ChIP), but targets RNA binding proteins. RIP precipitates a specific RNA binding protein (RBP) and associated RNA (mRNAs, non-coding RNAs, viral RNAs) that can be detected by real-time PCR, microarrays or sequencing. In addition to the immunoprecipitation methods, researchers sometimes fuse tags onto either the C-terminal or N-terminal of the protein of interest. This approach allows the same tag to be used for different proteins and eliminates the difficulty of generating different antibodies each time.
Creative Diagnostics can serve RIP technology coupled to either microarray (RIP-chip/rRIP-chip) or sequencing (RIP-seq) to identify RNAs that are bound by an RBP of interest.
RIP-chip/rRIP-chip: RIP-Chip can be used to identify subsets of RNAs that have related functions and are potentially co-regulated, as well as proteins that are associated with them in RNP complexes. A noteworthy special application of RIP-chip is recombinant RIP-chip (rRIP-chip). Our team can use recombinant RBPs of interest to probe an isolated total RNA sample and immunoselect the messenger-ribonucleoproteins using a specific anti-RBP antibody. The mRNA composition of the supernatant and/or immunoprecipitated fraction is analyzed using dual-color microarrays.
